Description of the Mastitis Test

Mastitis is the persistent, inflammatory reaction of the udder tissue due to physical trauma or microorganisms infections. Mastitis, a potentially fatal mammary gland infection, is the most common disease in dairy cattle worldwide. It is also the most costly disease to the dairy industry. Milk from reproductive animals suffering from mastitis has an increased somatic cell count.

Mastitis Test Principle

The somatic cells in the milk react with the hydrolysis substrate producing color formation and the buffer stabilizes the ongoing reaction by eliminating any interferences. The intensity of the color is directly proportional to the concentration of SCC in the milk. The advantages of the method are:
1. The current method has superior results in comparison to the esterase method
2. The test results have no interferences from the milk sample
3. Ideal tool for field dairy industry, instant colorimetric semiquantitative results in 1 minute. Makes obsolete the California Mastitis Test ( CMT)

Reagent Composition and Preparation
R1 – Buffer
R2 – Hydrolysis substrate

Stability: 24 months at RT. All the components of the kit are stable until the expiration date on the label.
Protect them from light and contaminations during their use. Do not use reagents over the expiration
date. Store the reagents at RT.
Samples: Raw milk from different reproductive animals

Procedure of the mastitis test

Add 2 drops of raw milk, add 2 drops of R1 and 1 drop of R2. Mix, incubate for 60 seconds and read the
color formed in comparison to the colorimetric index included in the kit for a semiquantitative
determination of total somatic cells count in the examined milk.
