Das Verfahren wurde gem\u00e4\u00df der ISO 1369:2003 (E) validiert und ist auf die Hemmung des mikrobiellen Wachstum basiert. Es wird in einem Mikrotiterplatten-Format, bei dem jede Vertiefung Agarmedium mit Geobacillus stearothermophilus enth\u00e4lt, zugegeben. Wenn die Platte bei 65 \u00b0 C inkubiert keimen die Sporen und die Zellen wachsen. Sie produzieren S\u00e4ure und \u00e4ndern so den Agar pH, was sich durch eine Farbe\u00e4nderung von blau auf gelb f\u00fchrt. Wenn Milchproben-Inhibitoren, in h\u00f6heren Konzentrationen als die Nachweisgrenze, enthalten, werden Mikroorganismen nicht wachsen.
