Der COVID-19 (SARS-CoV-2) -IgM\/IgG-ELISA dient zur qualitativen Bestimmung von Antik\u00f6rpern der IgG-Klasse gegen SARS-CoV-2 in Humanserum oder -plasma (Citrat, Heparin) zur Unterst\u00fctzung der Diagnose der COVID-19-Krankheit und stellt diese dar eine Erg\u00e4nzung zum direkten Nachweis von Krankheitserregern.<\/p>\n

Covid-Antigentest 19<\/span><\/span>\u00a0 Prinzip<\/h2>\n

Doppelantigen-Sandwich-Assay. Gesamtdauer des Assays: 70 Minuten Polystyrol-Mikrotiterplattenstreifen, die mit in Insektenzellen exprimierten rekombinanten SARS-CoV-2-Antigenen (rekombinantes N-Protein, S2-ECD und S1-RBD) vorbeschichtet sind. Die Serum- oder Plasmaprobe des Patienten wird hinzugef\u00fcgt, und w\u00e4hrend des ersten Inkubationsschritts werden die spezifischen SARS-CoV-2-Antik\u00f6rper in den Vertiefungen eingefangen, falls vorhanden. Die Mikrotiterplatten werden dann gewaschen, um ungebundene Serumproteine \u200b\u200bzu entfernen. Ein zweiter Satz rekombinanter Antigene (rekombinantes N-Protein, S2-ECD und S1-RBD), die an das Enzym Meerrettichperoxidase (HRP-Konjugat) konjugiert sind und dieselben Epitope wie die vorbeschichteten Antigene exprimieren, wird zugegeben und w\u00e4hrend der zweiten Inkubation werden sie an den eingefangenen Antik\u00f6rper binden. Die Mikrotiterplatten werden gewaschen, um ungebundenes Konjugat zu entfernen, und Chromogenl\u00f6sungen werden in die Vertiefungen gegeben. In Vertiefungen, die den „Sandwich“ -Immunokomplex Antigen-Antik\u00f6rper-Antigen (HRP) enthalten, werden die farblosen Chromogene durch das gebundene HRP-Konjugat zu einem blau gef\u00e4rbten Produkt hydrolysiert. Die blaue Farbe wird gelb, nachdem die Reaktion mit Schwefels\u00e4ure gestoppt wurde. Die Menge der Farbintensit\u00e4t kann gemessen werden und ist proportional zur Menge des in den Vertiefungen eingefangenen Antik\u00f6rpers bzw. zur Probe. Vertiefungen mit SARS-CoV-2-negativen Proben bleiben farblos.<\/p>\n
