Der COVID-19 (SARS-CoV-2) -IgG-ELISA dient zur qualitativen Bestimmung von Antik\u00f6rpern der IgG-Klasse gegen SARS-CoV-2 in Humanserum oder -plasma (Citrat, Heparin) zur Unterst\u00fctzung der Diagnose der COVID-19-Krankheit und stellt diese dar eine Erg\u00e4nzung zum direkten Nachweis von Krankheitserregern.<\/p>\n

Prinzip des Covid-19-Antigentests<\/span><\/span><\/h2>\n

Die qualitative immunenzymatische Bestimmung spezifischer Antik\u00f6rper basiert auf der ELISA-Technik (Enzyme-linked Immunosorbent Assay). Mikrotiterplatten werden mit spezifischen Antigenen beschichtet, um entsprechende Antik\u00f6rper der Probe zu binden. Nach dem Waschen der Vertiefungen, um alles ungebundene Probenmaterial zu entfernen, wird ein mit Meerrettichperoxidase (HRP) markiertes Konjugat zugegeben. Dieses Konjugat bindet an die eingefangenen Antik\u00f6rper. In einem zweiten Waschschritt wird ungebundenes Konjugat entfernt. Der durch das gebundene Konjugat gebildete Immunkomplex wird durch Zugabe von Tetramethylbenzidin (TMB) -Substrat sichtbar gemacht, das ein blaues Reaktionsprodukt ergibt. Die Intensit\u00e4t dieses Produkts ist proportional zur Menge spezifischer Antik\u00f6rper in der Probe. Schwefels\u00e4ure wird zugegeben, um die Reaktion zu stoppen. Dies erzeugt eine gelbe Endpunktfarbe. Die Absorption bei 450\/620 nm wird unter Verwendung eines ELISA-Mikrotiterplattenleseger\u00e4ts abgelesen.<\/p>\n
